GAITHERSBURG, Maryland (October 29, 2026) — BLA Regulatory (BLA) warmly congratulates its partner, NovaSpira Biotherapeutics, as the Investigational New Drug (IND) application for NS-342 successfully cleared review by the U.S. Food and Drug Administration (FDA), allowing the program to proceed to a Phase 1 clinical study in the United States. During the U.S. IND submission process, BLA served as NovaSpira’s external regulatory affairs team, leading the gap analysis, identifying potential regulatory risks, developing the submission strategy, preparing and reviewing the complete IND dossier, managing electronic Common Technical Document (eCTD) publishing and submission, and supporting the analysis of FDA review questions, document revisions, and response preparation after the application was accepted for review. This marks another milestone in the product’s global development, following authorization of its Clinical Trial Application (CTA) by South Korea’s Ministry of Food and Drug Safety (MFDS).

NS-342 is a next-generation GPC3 × 4-1BB-targeting bispecific molecule being developed for GPC3-positive solid tumors, with advanced hepatocellular carcinoma (HCC) as a core development focus. HCC is a highly aggressive malignancy, and treatment options remain limited for patients with advanced disease, representing a substantial unmet medical need.

Molecular Design Highlights: Selective Immune Activation at Tumor Sites to Reduce Systemic Toxicity

NS-342 features an innovative dual-target design intended to enable tumor antigen-dependent 4-1BB costimulation. By triggering T-cell activation only at GPC3-positive tumor sites, this approach is designed to avoid the systemic immune activation risks associated with conventional 4-1BB agonists. It has the potential to improve the therapeutic window while enhancing antitumor immune responses.

Preclinical data indicate that NS-342 selectively binds GPC3 on the surface of tumor cells, recruits and activates T cells within the tumor microenvironment, effectively lyses tumor cells, and increases tumor-infiltrating lymphocyte levels. It has demonstrated strong antitumor activity in animal models of liver cancer. The candidate has already received MFDS authorization for a Phase 1 clinical study in South Korea, with global clinical development advancing in parallel.

BLA’s Contribution: End-to-End Support for a Compliant FDA IND Submission

Bispecific antibodies are highly complex innovative biologics, and the FDA applies stringent review standards to their chemistry, manufacturing, and controls (CMC), nonclinical data, clinical protocols, risk assessments, and the logic and details of the submission dossier, creating a risk of deficiencies requiring correction or additional information. BLA’s integrated regulatory submission capabilities played a key role in the project’s efficient progress.

BLA was primarily responsible for the following activities during the project:

  1. In-Depth Evaluation of Complex Technical Issues

BLA’s senior experts conducted comprehensive assessments of key regulatory and technical questions involved in the bispecific antibody submission, drawing on the product’s mechanism of action, development data, and FDA regulatory requirements. They proposed targeted, actionable solutions to support critical project decisions.

  1. Regulatory Strategy and Submission Planning

BLA systematically reviewed the existing development data against relevant FDA regulations, guidelines, and the characteristics of NS-342, identified key regulatory concerns, and defined the documentation requirements and preparation strategy for each submission module, establishing a clear regulatory pathway for the U.S. IND.

  1. Preparation and Integration of Core IND Documents

BLA completed the integration, drafting, data alignment, and regulatory refinement of the full Common Technical Document (CTD) dossier, including CMC, nonclinical and clinical materials, risk assessments, and the Investigator’s Brochure, to ensure that the submission was coherent, rigorous, compliant, and ready for filing.

  1. Analysis of and Responses to FDA Review Questions

During the FDA review, BLA conducted regulatory and technical analyses of the questions raised by the Agency, developed tailored response strategies, drafted the relevant responses, and revised submission documents to support timely and accurate resolution of review concerns.

The successful FDA IND clearance for NS-342 represents another achievement for BLA in international regulatory submissions for innovative biologics. Looking ahead, BLA will continue to apply its cross-regional regulatory expertise to support innovative pharmaceutical companies in global clinical development and registration activities.
